Activation of PPARdelta by agonist L165041 in mice increases hepatic LPCAT3 mRNA abundance and LPCAT enzymatic activity, which is associated with increased incorporations of arachidonate into liver phosphatidylcholine and phosphatidylethanolamine. Liver X receptor (LXR) is the important transcription factor controlling LPCAT3 expression in liver tissue via an LXR response element (LXRE) in the proximal promoter region of the murine Lpcat3 gene. Adenovirus-mediated knockdown of PPARdelta in cultured hepatic cells and liver tissue reduced LPCAT3 mRNA levels, and exogenous overexpression of PPARdelta increases LPCAT3 mRNA expression.
